A reverse transcription loop-mediated isothermal amplification assay for the detection of citrus exocortis viroid in Australian citrus

چکیده

Abstract Citrus exocortis viroid (CEVd), the causal agent of exocortis, is a pathogen that thought to infect all citrus varieties, although it asymptomatic in most. Symptoms develop on susceptible rootstocks, resulting stunting and yield reduction. To aid detection management CEVd, rapid near-field assay was developed using reverse transcription loop-mediated isothermal amplification (RT-LAMP) for viroids nursery field trees. Over 240 CEVd sequences, including sequence variants from representative Australian isolates induce mild severe symptoms, were used design primers. The RT-LAMP successfully detected 1:1000 dilution (236 pg) plant total RNA indicating high sensitivity, also rapid, crude extractions. highly specific given there no cross-reactivity with other citrus-infecting pathogens. This new provides simple, robust, specific, sensitive method detect our knowledge, first any viroid.

Host-directed processing of Citrus exocortis viroid.

Prolonged infection of tomato hybrid (Lycopersicon esculentum x Lycopersicon peruvianum) by Citrus exocortis viroid (CEVd) resulted in viroid-like enlarged structures, detected by gel electrophoresis. This population included two new enlarged variants or D-variants, D-87 and D-76, and three transient species or D-forms, D-38, D-40 and D-43. Sequence analyses exposed a locus near the terminal re...

Reverse transcription loop-mediated isothermal amplification assay for rapid detection of Bovine Rotavirus

BACKGROUND Bovine rotavirus (BRV) infection is common in young calves. This viral infection causes acute diarrhea leading to death. Rapid identification of infected calves is essential to control BRV successfully. Therefore development of simple, highly specific, and sensitive detection method for BRV is needed. RESULTS A reverse transcription loop-mediated isothermal amplification (RT-LAMP) ...

Reverse transcription-loop-mediated isothermal amplification assay for rapid detection of hepatitis E virus.

The one-step single-tube betaine-free reverse transcription (RT)-loop-mediated isothermal amplification assay was developed for rapid diagnosis of hepatitis E virus. This assay amplified the target gene in less than 45 min (even as short as 20 min) under isothermal conditions at 63 degrees C, and the sensitivity of this assay was 100-fold greater than that of RT-PCR. This assay demonstrated a d...

A reverse transcriptase-loop mediated isothermal amplification assay (RT-LAMP) for rapid detection of bovine viral diarrhea virus 1 and 2

Bovine viral diarrhea virus (BVDV) is a pathogen that infects cattle, and is globally important. It causes substantial financial losses to the livestock industry. In the current study, a one-step reverse transcriptase-loop-mediated isothermal amplification (RT-LAMP) assay was set up for rapid and efficient detection of BVDV. For this purpose, four primers were designed to recognize six distinct...
